Mighty Rabbit Protocol

You have probably heard, or maybe even used, our revolutionary Express Rabbit protocol based on adjuvant IMMMACCEL-R™, capable of accelerating immune response and early switch to production of IgGs instead of IgMs. Lately we were working on increasing titers of specific antibodies in immune sera, and we are proud to present our new Mighty Rabbit protocol. Currently we offer it only for fusion proteins and other protein antigens.
                

For this protocol the protein is attached to a special branched molecule that also carries peptides stimulating the immune response.


What are the benefits of using Mighty Rabbit?

  • Additional activation of macrophages, Dendritic cells and T-helper cells results in better presentation of the antigen and stronger response
  • Immunization results in higher titers and stronger affinity antibodies
  • Serum has lower background since there are less antibodies of so-called non-defined repertoire and/or of low affinity
  • Smaller amounts of antigen can be used
  • No antibodies of undesired specificity appear since the attached construct has very low immunogenicity. Some anti-hapten antibodies against the core structure do not interfere with the main activity of sera (the structures of core never observed in nature)
  • in contrast to protein conjugated to a standard carrier molecule (BSA, KLH) the carrier does not compete with the protein for the "attention" of the immune system due to its low immunogenicity
  • The technology can be used in both Express, Express extended and standard long protocols
  • Titer of the serum obtained is 4-20 times higher as compared to the non-conjugated antigen (see the examples on the right)

Both examples below are not from preliminary laboratory tests but are actual customer projects:

Example 1

Antigen: fusion protein 30kD, low immunogenic.
Protocol used: Express Protocol, short, final bleed day 28.
Total amount of antigen used: 2 mg per protocol
Adjuvant Used: CFA/IFA, IMMACCEL-R
S-standard antigen mixed with CFA/IFA (white bars)
MR- modified antigen (orange bars)

Titers of 28 day sera were compared in ELISA on native fusion protein antigen:











As can be seen, titer of MR serum is approximately 8-16 times higher than that of serum obtained with normal protocol.


Strong activating signal to immune system results in not only higher titer, but also in higher specificity of serum. Specific and non-specific reaction is shown for standard protocol (SP) and Mighty Rabbit (MR) protocols. At much higher specific titer the serum also shows less background binding to BSA which was used as irrelevant antigen.

Example 2

Antigen:
fusion protein 15kD, immunogenic.
Protocol used: Express Protocol, short, final bleed day 28.
Total amount of antigen used: 2 mg per protocol
Adjuvant Used: CFA/IFA, IMMACCEL-R
